INTENDED USE

The Mouse anti-dsDNA IgG2b ELISA Kit is an immunoass aysuitable for quantifying or titering IgG2b antibody activity specificfor double-stranded DNA in serum or plasma. Other biologicalfluids, including tissue culture medium, may be validated for use.For research use only (RUO), not for diagnosis, cure orprevention of the disease.

GENERAL INFORMATION

Antibodies reactive with autologous nuclear components, such asDNA and histones, can represent an autoimmune basis forpathological conditions such as systemic lupus erythematos is(SLE) in humans, and in mice homozygous for thelymphoproliferation spontaneous mutation (Faslpr), a systemicautoimmunity with massive lymphadenopathy associated withproliferation of aberrant T cells, arthritis and immune complexglomerulonephritis. These conditions include elevated levels ofanti-dsDNA and other anti-nuclear antibodies (ANA) which oftenincrease as the animal ages. Also, the expanded use in the drugindustry of biological modifiers has been associated withproduction of autoantibodies, of which mice, and possibly alsoother hosts such as humans and monkeys, are susceptible. Aprototype disease in mice is lupus caused by the drugminocycline, with elevated anti-dsDNA among otherautoantibodies and pathological conditions.Recent investigations have focused on the role of innate immunemechanisms, including Toll-like receptors (TLRs) and TH2immunity, responding to the damage-associated molecularpatterns of dying cells, as underlying cause of the anti-ds DNAtype autoimmunity; these may be induced by drugs, includingvaccines and adjuvants, with aging, or with other conditions.

Specificity

Purified dsDNA is used to coat the microwells; thus the assay isspecific for antibodies directed to dsDNA. The anti-Mouse IgG2bHRP conjugate reacts specifically with mouse IgG2b subclassantibodies that bind to dsDNA on the plate. IgA, IgM, IgE andother IgG subclass antibodies would not be measured abovebackground signals.

Assay Sensitivity

The dsDNA coating level, HRP conjugate concentration and LowNSB Sample Diluent are optimized to differentiate anti-ds DNAIgG2b from background (non-antibody) signal with mouse serumsamples diluted 1:100.
